M9 beta-glucosidase transformant plates — unexpected growth pattern

M9 beta-glucosidase-transformant plates were read.

Result did not match prediction. Cellobiose-only plate showed the most growth, glucose-only showed less, and cellobiose+glucose combined showed only a single colony — the prediction was that the combined plate should be comparable to or better than cellobiose alone. Separately, an M9 plate with DH5α and no supplement (expected to show no growth) instead showed clean colonies, unlike every previous observation on M9 plates without supplement.

Both results are flagged as inconsistent with expectations and unresolved as of this entry.

Beta-glucosidase-transformant glycerol stock was recovered by scraping ice from the top of the frozen stock into 10 mL LB + 34 µg/mL chloramphenicol, and streaked onto a standard LB + chloramphenicol plate (from the front fridge stock).
