Picked a single Top10 + CxnA plasmid colony (fresh LB + chloramphenicol plate, or scraped from glycerol stock), inoculated into 5 mL LB + 40 µg/mL chloramphenicol, and grown overnight at 37 °C, 200 rpm (14–18 h) — feeder culture for the M9 growth-curve experiment run the next day.
