Transformed BL21 with the beta-glucosidase plasmid (positive-control strain for downstream activity assays): 50 µl BL21 + 2 µl plasmid, 30 min on ice, heat shock 42 °C for 40s (deviation: 1 min at 37 °C shaking noted), 40s on ice, 37 °C for 20 min (deviation: SOC was forgotten at this step and added afterwards — 1 mL SOC, then 50 min at 37 °C), plated 100 µl onto LB + chloramphenicol.
Cells were also spun down (13k, 1 min), supernatant removed, and the pellet resuspended in 100 µl SOC for a concentrated plating — entry cuts off before the outcome of this step was recorded.
