Cellobiose selection (Selection 2)
Grew Project A and Project B transformants (survivors of chloramphenicol Selection 1) on cellobiose plates. Colonies surviving this selection have acquired activity on cellobiose as sole carbon source — the primary functional screen.
Miniprep
Miniprepped surviving mutant CxnA from both Project A and Project B. Standard alkaline lysis/spin column protocol.
Sequencing
Sent miniprep samples for Sanger sequencing to characterise mutations in selected survivors.
