Inoculated a single colony of Top10 E. coli (~5 µl) into 10 ml LB, cultured overnight at 37 °C, 200 rpm to begin making chemically competent cells (~2 hours active work).
Result: Culture not used — mistakes made during preparation. Attempt abandoned.
