The results of PCR showed the products of homologous recombination were not contaminated by wild-type phage, but might be contaminated by kanamycin section. However, we might have constructed SP successfully; the sequence results showed the products were M13 phage backbone templates, and we surely constructed SP successfully, which matched sequence results completely; no phage plaques existed on the plates that day.

2022/4/24 | Y.R. | Two layer plating method: 150 μL bacterial solution+ 10 μL phage solution+ 22 μL bluegal coloring solution+ 1 ml 2xYT liquid media(make 4 dilution gradients for phage solution: original concentration, 102 times diluted, 104 times diluted, 106 times diluted). | Phage plaques appeared. |

PCR products of all 8 phage plaques produced bright bands after electrophoresis by using the primer of T7; PCR products of cloudy bacterial solution produced no bright bands after electrophoresis by using the primer of T7; PCR products of 2 phage plaques on the plate where the phage plaques seemed to appeared on produced one bright strip and the other was not brighr after electrophoresis by using the primer of T7; phage solution whose strip was bright was taken into the 2xYT liquid media in the test tubes and put them in the biological shaker; all the results showed a obvious problem: in this validation, PCR products of wild-type phage produce a strip at the position of 3300 bp by using the primer of T7. The following analysis showed this results was not trustworthy, and the results might be caused by primer contamination.
2022/4/25 | Z.Y.H. | Pick single colonies of bacteria S2060 into the tubes and put them in the biological shaker. | Make competent cells. |

However, the PCR products of kanamycin gene produced strips at the position of over 1000 bp by using the primer GOIF&R; the results of experiment C prodcued no strips which meant the phage was wild type.

L.W.R., L.J. | PCR: T7-1; operate electrophoresis and gel extraction(M13, T7-1). | PCR products of M13 phage backbones produced diffused strips; PCR products of T7-1 produced one strip, but the concentration of T7-1 was too low.

The results showed only kanamycin gene section can be amplified by using the phage solution after propagation and primers GOIF&R; PCR products of phage plaques appearing on the plates painted with homologous recombination products produced no strips by using the primer T7 which meant only wild-type phage grew on the plates.
